Gene expression pattern of digestive and antioxidant enzymes during the larval development of reared Atlantic bluefin tuna (ABFT), Thunnus thynnus L. ArchiMer

The aim of this study was to determine whether mortality observed during the larval development of reared bluefin tuna (Thunnus thynnus) could be related to improper expression profiles of key genes involved in digestive or antioxidant response capabilities. Tuna larvae were sampled at hatching, 2, 5, 10, 15 and 20 dph (days post hatching) for the relative quantification of transcripts encoded by genes involved in digestive [trypsinogen 1 (TRYP1), alpha-amylase (AMY), aminopeptidase N (ANPEP)] and antioxidant [catalase (CAT)] functions. Heat shock protein 90 of Bonamia ostreae: characterization and possible correlation with infection of the flat oyster, Ostrea edulis ArchiMer

In this study we described the cytosolic HSP90 of Bonamia ostreae, an intracellular parasite of Ostrea edulis haemocytes. The complete open reading frame was assembled by Rapid Amplification cDNA Ends reactions on cDNA of B. ostreae infected haemocytes. HSP90 amplification was corroborated in infected oysters and B. ostreae purified cells. The functionality of the HSP90, studied by inhibitory assays with radicicol, suggests that this protein may play a role in haemocyte invasion. Our results inform the molecular basis that governs B. ostreae-O. edulis interactions.

Identification of multi-drug resistance gene (MDR1) in equine ileum Ciência Rural

P-glycoprotein (P-gp) is a membrane transporter encoded in the Multi-drug Resistance (MDR1) gene expressed in several normal tissues and over expressed in tumor cells. P-gp was already identified in different species but not yet in equine. MDR1 gene and P-gp are able to interfere with bioavailability and disposition of several drugs, altering pharmacokinetic and pharmacodinamic of drugs. The presence of the MDR1 and P-gp in the central nervous system blocks the entry of certain drugs in this tissue and reduces drug absorption and enhances drug elimination when P-gp and MDR1 are presented in the gastrointestinal tract. The history of French oyster culture shows very well how fragile this farming is to stock overexploitation and diseases. The production of flat oyster, Ostrea edulis, fell dramatically after the appearance of two parasitic diseases: arteiliosis and bonamiosis. The curative means against bonamiosis (Bonamia ostreae) are restricted and imply a good knowledge of the parasite's life cycle and of its interaction with its host. Gene therapy is an active field that has progressed rapidly into clinical trials in a relatively short time. The key to success for any gene therapy strategy is to design a vector able to serve as a safe and efficient gene delivery vehicle. This has encouraged the development of nonviral DNA-mediated gene transfer techniques such as liposomes. Many liposome-based DNA delivery systems have been described, including molecular components for targeting given cell surface receptors or for escaping from the lysosomal compartment. Another recent technology using cationic lipids has been evaluated and has generated substantial interest in this approach to gene transfer.

Summer mortality of Crassostrea gigas is the result of a complex interaction between oysters, their environment, and pathogens. A high heritability was estimated for resistance to summer mortality, which provided an opportunity to develop lines of oysters that were resistant (R) or susceptible (S) to summer mortality. Previous genome-wide expression profiling study of R and S oyster gonads highlighted reproduction and antioxidant defense as constitutive pathways that operate differentially between these two lines. Here, we show that signaling in innate immunity also operates differentially between these lines, and we hypothesize that this is at the main determinant of their difference in survival in the field. To investigate the control at the mRNA level of glycogen metabolism in the cupped oyster Crassostrea gigas, we report in the present paper the cloning and characterization of glycogen phosphorylase and synthase cDNAs (Cg-GPH and Cg-GYS, respectively, transcripts of main enzymes for glycogen use and storage), and their first expression profiles depending on oyster tissues and seasons. A strong expression of both genes was observed in the labial palps and the gonad in accordance with specific cells located in both tissues and ability to store glucose. Molecular Cloning and Sequencing of AlkalophilicCellulosimicrobium cellulans CKMX1 Xylanase Gene Isolated from Mushroom Compost and Characterization of the Gene Product BABT

ABSTRACT A xylanolytic bacterium was isolated from mushroom compost by using enrichment technique. Results from the metabolic fingerprinting, whole-cell fatty acids methyl ester analysis and 16S rDNA sequencing suggested the bacterium to be Cellulosimicrobium cellulans CKMX1. Due to the xylanolytic activity of this bacterium, isolation and characterization of the xylanase gene were attempted. A distinct fragment of about 1671 bp was successfully amplified using PCR and cloned into Escherichia coli DH5α. A BLAST search confirmed that the DNA sequence from the amplified fragment was endo-1, 4-beta-xylanase, which was a member of glycoside hydrolase family 11. Multi-drug resistance gene (MDR1) and opioid analgesia in horses Ciência Rural

Opioid absorption in the intestinal tract as well as its effects in the central nervous system is modulated by the P-glycoprotein (P-gp) encoded in the Multi-drug Resistance gene (MDR1) also named ATP-binding cassete, subfamily B, member 1 (ABCB1). This MDR1 gene acts as a selective pump. The expression of this protein in humans and rodents inhibits cellular uptake of substrate opioids. The presence of the intestinal iso-enzyme CYP3A4 associated with MDR1 gene decreases the opioid analgesic activity due to an increase in intestinal metabolism, with a predicted intestinal first pass extraction around 20% which significantly influences the oral availability of opioids. Grey level co-occurrence matrix analysis (GLCM) is a well-known mathematical method for quantification of cell and tissue textural properties, such as homogeneity, complexity and level of disorder. Isolates of Salmonella sp (104) from poultry samples were isolated and serotyped where eleven were identified as Salmonella Typhimurium (ST). ST isolates were phenotypically tested by disk diffusion and minimum inhibitory concentration (MIC). Four genes related to enrofloxacin (GyrA, GyrB, ParC and ParE), two to gentamicin (AadA and AadB) and two to ceftiofur (BlaCMY-2 and AmpC) resistance were searched by PCR. Our results showed ST resistance to all three antibiotics tested (18.1% for ceftiofur, 45.4% for gentamicin, and 18.1% for enrofloxacin) according to the diffusion test. Bacillus thuringiensis harbors genes encoding Cry proteins found in chromosomes or plasmids of different sizes (4-150 Mb). Although the smaller plasmids are more abundant in B. thuringiensis, their specific function is unknown.
